Anyone who's ever had a nice brisk bike ride through a snowstorm is likely to cringe a bit at the above image. Though the gentlemen pictured is looking forward to the summer season for other reasons -- that's when the Recon heads-up display he's testing is set to get a bigger reveal. In the meantime, we're stuck staring at the above image of HUD-equipped sunglasses and wondering precisely what it all means -- aside, of course, for the company's move off the ski slopes.

Stressed proteins can cause blood clots for hours

Mar. 5, 2013 ? New research from Rice University, Baylor College of Medicine (BCM) and the Puget Sound Blood Center (PSBC) has revealed how stresses of flow in the small blood vessels of the heart and brain could cause a common protein to change shape and form dangerous blood clots. The scientists were surprised to find that the proteins could remain in the dangerous, clot-initiating shape for up to five hours before returning to their normal, healthy shape.

The study -- the first of its kind -- focused on a protein called von Willebrand factor, or VWF, a key player in clot formation. A team led by Rice physicist Ching-Hwa Kiang found that "shear" forces, like those found in small arteries of patients with atherosclerosis, cause snippets of nonclotting VWF to change into a clot-forming shape for hours at a time. The finding appears online this week in Physical Review Letters.

"When I first heard what Dr. Kiang's team had found, I was shocked," said blood platelet expert Dr. Joel Moake, a study co-author who holds joint appointments at Rice and BCM. Moake, whose research group was the first to describe how high shear stress could cause platelets to stick to VWF, said, "I had thought that the condition might last for such a short time that it would be unmeasurable. No one expected to find that this condition would persist for hours. This has profound clinical implications."

Kiang, associate professor of physics and astronomy and of bioengineering, studies the forces involved in protein folding. Proteins are the workhorses of biology. Tens of thousands are produced each second in every living cell, and each of these folds into a characteristic shape within moments of its creation. Despite its ubiquity, protein folding is an immensely complex process that is shrouded in mystery.

Kiang is a pioneer in the use of atomic force microscopes (AFM) to shed light on the fundamental physical processes involved in protein folding. The AFM has a tiny needle with a tip measuring just a few atoms across. The needle is suspended from a tiny arm that bobs up and down over a surface. Kiang's team uses the bobbing needle to grab and pull apart individual protein molecules. By stretching these like rubber bands, her team has shown it can measure the precise physical forces that hold them in their folded shape.

"In this study, we did more than just measure the forces; we used those measurements to see what state the molecule was in," Kiang said. "In this way, we were able to study the dynamics of the molecule, to see how it changed over a period of time."

Moake, a senior research scientist in bioengineering at Rice and professor of medicine at BCM, said the work is vitally important because it helps explain the workings of VWF.

"VWF is synthesized in the cells that line the walls of blood vessels, and it's stored there until the cells get signals that the vessels are in danger of injury," Moake said. "In response to those stimuli, the cell secretes VWF. It's a long protein, and one end remains anchored to the cell while the rest unfurls from the wall like a streamer."

The act of unfurling makes VWF sticky for platelets, and that begins the process of hemostasis, which prevents people from bleeding to death when blood vessels are damaged by cuts and wounds.

"The body recognizes when clotting must stop -- when there are too many strings, too much sticking, too many platelet clumps -- and it uses an enzyme to clip the long VWF strings," Moake explained. "First, it makes large, soluble versions of the strings that remain somewhat sticky, and then these large soluble portions of VWF are reduced into smaller subunits of VWF that circulate in the plasma."

Under normal conditions, these circulating subunits, which are called PVWF, fold into compact shapes and cease to be sticky to platelets. However, previous research had shown that a type of physical stress called "shear" -- which can arise in partially occluded arterial blood vessels with high flow rates -- could cause PVWF to become sticky to platelets.

"That's all we knew," Moake said. "We didn't know how the conformation of the PVWF protein changed. That is why Dr. Kiang's research is so important and makes it more likely that therapeutic interventions can be more rationally designed."

To study the problem, Kiang's lab worked closely with Moake's team at Rice's BioScience Research Collaborative and with researchers from the laboratory of co-author Jing-fei Dong, formerly of BCM and now at PSBC in Seattle. Moake's and Dong's groups prepared samples of PVWF, subjecting some to the shear stresses known to induce clot formation. Kiang's team used AFMs to test the samples. Through a combination of experiments and deductive reasoning, her team determined exactly which portion of PVWF changed its conformation during shear stress. They also determined how long the protein remained partially unfurled before relaxing into its natural shape.

"The next step will be to design new experiments that allow us to monitor the proteins as they bind to platelets and initiate clot formation," Kiang said. "That will tell us even more about the physical properties of the proteins and provide more clues about potential therapies."

The research was supported by the National Institutes of Health, the National Science Foundation, the Alliance for NanoHealth, the Welch Foundation, the Mary R. Gibson Foundation and the Everett Hinkson Fund. Study co-authors include Rice graduate students Sithara Wijeratne and Eric Frey, former Rice graduate student Eric Botello, BCM researchers Hui-Chun Yeh and Angela Bergeron, Rice undergraduate Jay Patel, PSBC's Zhou Zhou and Rice senior research technicians Leticia Nolasco and Nancy Turner.

Will Community colleges' cash crunch threaten Obama's retraining plan?

PALATINE, Illinois (Reuters) - Some schools rally around a football team. Harper College has rallied around a number.

When President Barack Obama called for 5 million more community-college graduates by the end of the decade to boost U.S. competitiveness, this commuter school 30 miles northwest of Chicago figured out it would need to produce 10,604 additional graduates to do its part.

It won't be able to count on much government help.

Even as Harper and many of the nation's 1,200 other two-year community colleges try to deliver on Obama's vision of a revitalized manufacturing sector and a better-skilled work force, support from the federal and state governments is eroding.

"I've been in this business for 42 years. I've never seen anything like this - the pressure on the business model, the pressure on the whole institution of higher education," Harper College President Ken Ender said in an interview.

The problem is especially acute in Obama's home state of Illinois, where decades of mismanagement have led to a fiscal crisis that is squeezing state spending on education.

State funding covered 5.8 percent of Harper's budget last year - a record low and down from 13.3 percent a decade ago. Next year is likely to be worse, Ender says.

It reflects a trend in Illinois and across the nation that threatens to make education less affordable to millions of students at two-year colleges, which can provide a start for those working toward a four-year bachelor's degree or enable older workers to learn the skills needed for new careers.

Since he took office in 2009, Obama has lavished attention on two-year schools; last year he visited 10 community college campuses. Last month, he flew to North Carolina after his State of the Union speech to Congress to highlight a manufacturer that was working with a local school to train workers.

The attention hasn't translated into more financial support.

Obama's efforts to expand spending on education, job training and other elements of what he has termed an "innovation agenda" have fallen flat as Republicans have pushed for spending cuts to rein in trillion-dollar budget deficits.

Deficit-reduction efforts, including the across-the-board "sequester" cuts that kicked in on Friday, have fallen largely on the discretionary part of the federal budget that funds these areas, rather than the health programs that are projected to drive up the nation's debt in the decades to come.

Obama won $2 billion for community-college training programs when his Democrats controlled Congress in 2009, but last year's $8 billion proposal went nowhere.

He has had more success expanding the Pell grants that help nearly half of all community-college students pay tuition. But budget pressures have led to cutbacks in that program as well.

Meanwhile, state and local governments have slashed their higher-education spending during recession-induced budget crises. As a result, students have been forced to pay more for their education.

According to the American Institutes for Research, the average annual community-college tuition rose by 41 percent, to $3,269, from 2000 to 2010. The average subsidy per student received by community colleges fell by 24 percent during that period, to $6,223 per year.

Public funding for four-year public colleges has ebbed as well, but the trend has been most pronounced at the community-college level, the nonprofit group found.

As tuition has climbed, students have taken on more debt, and there are signs that many may not be able to keep up. Student debt has nearly tripled since 2004 and loan delinquency rates have been climbing, the Federal Reserve of New York says.

Businesses that consider community colleges to be an essential source of skilled workers view the trend with alarm.

"We need to continue to have government funding for these vocational schools," Natalie Schilling, vice president of human resources at aluminum giant Alcoa Inc, said during a recent panel discussion in Washington.

Like other manufacturers, Alcoa relies on community colleges to replace its aging work force and produce students who can operate the computer-controlled machines that are increasingly common in U.S. factories.

"We can't do it alone - it's got to be a shared responsibility among the public and private sector," Schilling said.

NEW FINANCIAL REALITY

In Illinois, the state's fiscal crisis has put its pension fund $98 billion in the red and dropped its bond rating to one of the lowest among U.S. states.

Since fiscal 2008, pension costs have grown from 8 percent to 20 percent of state revenue, according to the State Budget Crisis Task Force, an independent research group co-chaired by Paul Volcker, a former chairman of the U.S. Federal Reserve. State spending on the government-run Medicaid health program for the poor and disabled has doubled since 2000.

Meanwhile, state spending on higher education in Illinois has dropped by an inflation-adjusted average of 3.3 percent per year over the past 10 years.

Community colleges in less affluent areas have been hit especially hard in a pattern that could undermine some of Obama's efforts to help the nation's poor work their way into the middle class.

The president of Southeastern Illinois College in rural Harrisburg worries that continued tuition increases could drive students away as state contributions shrink from 45 percent of the college's budget to 35 percent in the next several years.

"At some point, depending on your demographic and your tuition charge, you're going to push students off the cliff," Jonah Rice said. "We're doing the best we can, but it's tough when the rubber band is stretched."

Nestled in Chicago's high-cost suburbs, Harper College has been somewhat shielded from the state's fiscal crisis because it can rely on local property taxes for about half of its operating costs. Schools in areas where property values are low, like Southeastern, are more exposed.

Budget woes, however, have not stopped Harper from forging ahead on its goal to boost graduation rates, which have jumped in the past two years.

Education groups point to Harper as a model for how to train a generation of tech-savvy manufacturing workers, and Republican presidential candidate Mitt Romney praised its manufacturing program during a campaign stop last year.

"To say we can't do things because there's no money is frankly unacceptable," Ender said. "There is no money. So that means we're not ever going to do anything?"

Harper has set up an outreach program with local high schools to try to reduce the number of incoming Harper students who need to take remedial math and reading courses.

Guidance counselors work with students to keep them on track toward their goals, whether that is a welding certificate or a transfer to a four-year college.

'WHEN IS THIS GOING TO STOP?'

Harper also has set up a paid internship program with local manufacturers that allows students to earn money while they learn - an important consideration for many.

"When I found out it was affordable, I jumped in. Every other type of training I've been looking at has not been affordable," said Peter McManus, 47, who is interning at a nearby packaging plant as part of the school's advanced-manufacturing program.

Like many others at Harper, McManus doesn't fit the mold of a traditional college student.

Seeking a career change after his job as a technical theater director was eliminated, McManus learned about Harper's manufacturing program through his teenage son. When he's not in class, McManus works at Transparent Container Company Inc. as he pursues a degree in automation.

"I grew up a mechanic's son and a tinkerer. To me, this is just another lab to tinker in," he said as he gazed at Transparent's $500,000 machines that stamp out plastic packaging.

Harper has gotten some recent government help: The school won a $13 million grant from the U.S. Labor Department last fall to expand its manufacturing internship to other community colleges in the region.

Nevertheless, Harper has been forced to raise tuition rates steadily as state funding has receded. Full-time students will pay $3,735 in the next academic year, an inflation-adjusted increase of 16 percent since the 2008 school year.

The college aims to keep tuition increases in line with inflation, and Ender says the school remains affordable for most.

Students who live with their parents should be able to cover tuition by working full-time in the summer and part-time during the school year, while less affluent students can apply for Pell grants to cover tuition, he says.

Still, 25 percent of students who drop out of Harper do so for financial reasons, said Maria Moten, the school's enrollment dean. Student leaders say they're worried that further tuition hikes will squeeze out more students.

"Most people at community colleges are working full-time jobs and coming to school. They're paying rent; some are paying for children," said Scott Leitzow, a member of the school's student senate. "When is this going to stop? I don't think it ever will."

Cardinals say will not be rushed into electing new pope

VATICAN CITY (Reuters) - Catholic cardinals said on Tuesday they wanted time to get to know each before choosing the next pope and meanwhile would seek more information on a secret report on alleged corruption in the Vatican.

Nearly 150 cardinals held a second day of preliminary meetings, known as "general congregations", to sketch a profile for the next pope following the shock abdication of Pope Benedict last month.

Under Church law they have until March 20 to start a conclave to choose a new pope from among 115 of them who are under the age of 80, but they can decide to start it earlier.

While many observers had expected the conclave to begin as early as this Sunday, there have been increasing indications that the cardinals may need more time to ponder who among them might be best to lead a church beset by crises.

"Many cardinals are concerned that if there is not enough time spent in the general congregations that once we get into the conclave it could drag on," Boston Cardinal Sean O'Malley said.

"I think the preference would be to have enough discussions previous that when people go to the conclave they already have a sort of pretty good idea of who they are going to vote for at that point," he told a news conference.

The preliminary meetings are taking place as the crisis involving sexual abuse of children by priests and inappropriate behavior among adult clerics continues to haunt the church and has rarely been out of the headlines.

One elector - Cardinal Keith O'Brien - quit as Edinburgh archbishop last week and pulled out of attending the conclave because of accusations that he behaved inappropriately with priests and seminarians in the past.

He at first denied the allegations but on Sunday issued a statement apologizing that "my sexual conduct has fallen below the standards expected of me as a priest, archbishop and cardinal".

Cardinals will be using the daily preliminary meetings, which began on Monday, to get to know each other and decide when to start the closed-door conclave to choose a man to lead the 1.2 billion-member Church at one of most difficult periods in its history.

"This is the most important decision that some of us will ever make and we need to give it the time that's necessary," O'Malley said.

Cardinal Daniel DiNardo of Galveston-Houston, said: "It takes as long as it takes ... no one wants to rush this if it can't be rushed."

The prelates, however, have said that if at all possible they would like to be home for Palm Sunday, which falls on March 24, meaning that a new pope would have to be elected and installed at a separate ceremony before then.

SISTINE CHAPEL CLOSED

There was no indication when the cardinals would decide on the starting date of the conclave.

The Sistine Chapel, where it will be held, was closed to the public on Tuesday to allow workmen to prepare the Michelangelo-frescoed room for the gathering, disappointing many tourists who had come to Rome to see it.

For the second consecutive day at the closed-door preliminary meetings the cardinals discussed the Vatican's often dysfunctional central administration, known as the curia, and a secret report on last year's so-called "Vatileaks" scandal.

The leaks scandal led to the arrest of Paolo Gabriele, the pope's butler, who was convicted of stealing personal papal documents and leaking them to the media. The documents alleged corruption and infighting over the running of its bank.

Benedict, who later pardoned Gabriele, decided to make the report available only to his successor but three elderly cardinals who wrote it are attending the preliminary meetings and are expected to brief their brother cardinals.

"We want to know and learn as much as we can relative to governance in the Church," said DiNardo.

Vatican officials have said the report's authors could "use their discernment" to give any necessary guidance to fellow cardinals without violating their pact of secrecy about its specific contents.

"I think the cardinals feel confident that we will get all the information that we need for our deliberations. It doesn't necessarily mean that the report will be shared with us but if there is anything germane that we need to know about I am sure that will be given," O'Malley said.

Four-year-old unhurt after falling from third story window

?I fall ? really, really far,? 4-year-old Dylan Hayes said in an interview after miraculously surviving a three story drop during which the child somersaulted twice before landing on his feet.

Local Denver CBS affiliate KCNC reports that young Dylan was standing on the living room couch trying to talk to his downstairs neighbors when he fell through.

?I was terrified,? his mother Jessica Hayes told the station. ?I didn?t know what I was going to see when I got down to the ground floor.?

She had moved the couch next to the window Dylan fell through because she was shampooing the living room carpet.

?I didn?t know what I was going to go down to, like a child who was just lying there, or a child with blood coming out of him,? Hayes said during what must have been the longest trip down that flight of stairs in her life.

Despite landing on his feet, Dylan was taken to a local hospital and kept under observation for 20 hours. Doctors placed a brace on his neck as a precaution but said he appears to not have suffered any serious physical injuries. Even more amazing, Dylan landed on a bed of rocks.

The ordeal led to one news outlet referring to Dylan as a ?superboy,? for his miraculous luck.

?There had to have been angels watching over him,? Dylan?s grandmother Kelli Hayes told the station.

For her part, Jessica Hayes said the incident has reminded her to never make assumptions about her child?s safety. She?s also moved out of the apartment and is looking for a new home on the ground floor.

?It?s gotten easier since it happened two weeks ago to not blame myself,? she said. ?I wish the danger would have crossed my mind, which it didn?t.?

Cancer vaccines self-sabotage, channel immune attack to injection site

Mar. 3, 2013 ? Cancer vaccines that attempt to stimulate an immune system assault fail because the killer T cells aimed at tumors instead find the vaccination site a more inviting target, scientists at The University of Texas MD Anderson Cancer Center report in Nature Medicine.

A common substance used in many cancer vaccines to boost immune attack betrays the cause by facilitating a buildup of T cells at the vaccination site, which then summon more T cells to help with the perceived threat.

"Vaccines stimulate production of T cells primed to attack the target cancer, and there are many T cells in the bloodstream after vaccination. We found that only a few get to the tumor while many more are stuck at or double back to the vaccination site," said senior author Willem Overwijk, Ph.D., in MD Anderson's Department of Melanoma Medical Oncology.

The result: largely unscathed tumors while an overstimulated immune response can cause lesions at the injection site. The team found that a major culprit in this failure is incomplete Freund's adjuvant (IFA), a mineral oil-based adjuvant included in many vaccines to stoke the immune response.

"IFA sticks around the vaccination site for up to three months, along with the antigen designed to trigger immunity against the tumor," Overwijk said. "T cells keep attacking and secreting chemokines to call for reinforcements. But it's an unkillable target; T cells can't kill mineral oil."

Eventually, the T cells die. "The vaccination site increasingly resembles a viral infection, with lots of damaged tissue and antigens," Overwijk said.

Switch from IFA to saline adjuvant reverses effect

"Switching to a saline-based adjuvant in a melanoma vaccine reversed the T cell effect in mice," Overwijk said, "Major accumulations of T cells gathered in tumors, shrinking them, with minimal T cell activity at the vaccination site."

Peptide antigens are available for almost all types of cancer, Overwijk said. A saline adjuvant could change the poor performance of cancer vaccines.

A clinical trial of the concept is expected to open later this year led by Craig Singluff Jr., M.D., professor of surgery at the University of Virginia Medical School, and Patrick Hwu, M.D., chair of MD Anderson's Department of Melanoma Medical Oncology.

Overwijk and colleagues noted 98 federally approved U.S. clinical trials of vaccines against a variety of cancers have almost all failed, while another 37 trials are open, enrolling patients. The U.S. Food and Drug Administration has approved only one therapeutic vaccine, for treatment of prostate cancer, out of all of those trials.

"Our group and many other researchers have been trying for years to improve the performance of cancer vaccines, to no avail," Overwijk said. "People kept trying because of these beguiling T cell levels in the blood. But our data suggest that the very nature of IFA-based vaccines may make it almost impossible for them to work well."

In past experiments and clinical trials, tumors were rarely examined for evidence of T cell penetration. In people, they are often inoperable, and there was no indication that it needed to be done. "But a few researchers did analyze human tumors for T cell infiltration and largely found what we found in our mouse experiments," he said.

Mouse studies reveal vaccine self-sabotage

The team studied the fate of melanoma-specific CD8-positive T cells after vaccination with the gp100 peptide with and without IFA.

Both vaccines increased levels of the desired T cells in the blood, but with IFA, the T cells dropped to nearly undetectable levels after three weeks and did not rebound even with an engineered virus-based booster. The vaccine-lacking IFA produced similar peak amounts of the T cells, a response that persisted over time.

The research team fluorescently tagged T cells in the mouse model to see where they went.

* Mice without IFA had the bulk of T cells light up in their tumors with minimal presence at the vaccination site.

* T cells built up at the injection site in mice that received IFA-based vaccine, with a tiny showing in the tumor.

Response duration was tested in gp100/IFA and control IFA vaccines. The antigen/IFA combination gathered and persisted at the vaccination site, where it could still stimulate the proliferation of injected T cells 96 days after vaccination.

A separate set of experiments showed the antigen/IFA-driven T cells were forced to kill themselves at the vaccination site by a variety of cell suicide-inducing proteins.

Reducing vaccine depots at injection site

Overwijk and colleagues inferred that a possible answer to the problem was to reduce the size and persistence of vaccine "depots" at the injection site.

They tested a vaccine based on a saline solution instead of IFA and found that antigens cleared more quickly but did not spark the desired T cell response. A combination of three stimulatory molecules (covax) was added to the saline/peptide vaccine, producing a strong T cell response. IFA/peptide vaccine produced a strong T cell response but also stronger post-peak T cell suicide.

A comparison of saline/peptide/covax vs. IFA/peptide/covax showed the saline version caused T cells to home to the tumor and destroy them, while the IFA version focused T cells at the vaccination site, killing normal tissue and inducing chemokines that damaged and killed T cells.

"IFA-based vaccination sites essentially outcompete tumor sites for T cell recognition and accumulation, chemokine production and tissue damage," Overwijk said. "It's an engineering flaw in those vaccines that we didn't appreciate until now. Fortunately, our results also directly instruct us how to design new, more powerful vaccine formulas for treating people with cancer."
